Does this VNA measure S21 (transmission)?

This kit is designed for S11 reflection measurement only; it uses a directional bridge. For transmission, you'd need a second bridge and RF switch.

What frequency range does it cover and how accurate is it?

The AD9833 generates 1 kHz to 40 kHz (usable up to about 100 kHz with roll-off). Accuracy depends on your calibration, but with a known load you can achieve approximately 5% impedance error.

Do I need an RF-shielded enclosure?

For best results, place the bridge and op-amp circuits on a ground plane; the kit includes prototyping boards to solder your own shielding if needed.
